📋 Regulatory Action / Label Change

Casgevy (exagamglogene autotemcel) was previously approved for patients aged 12 years and older with sickle cell disease (SCD) or transfusion-dependent beta thalassemia (TDT). The FDA has now expanded eligibility to children as young as 2 years following positive pediatric phase 3 results.


Clinical Considerations

  • All 8 efficacy-evaluable pediatric SCD patients achieved 12 months free from severe vaso-occlusive crises.
  • In pediatric TDT, 8 of 9 efficacy-evaluable patients achieved 12-month transfusion independence.
  • Approval for children aged 2 to less than 5 years relied on extrapolated clinical and product-characteristic data.
  • Grade 3-4 adverse events included mucositis, febrile neutropenia, cytopenias, and decreased appetite.
